NIH3T3 cells transfected with inducible expression vectors harboring the protein tyrosine phosphatase alpha (PTPalpha) gene were used as a model to study the mechanism of tumor formation, by which the changes of phenotypes in inducible cells were studied. When NIH3T3 cells transfected with PTPalpha were induced for 24 hours, the expression levels of PTPalpha in inducible cells was higher than those in uninducible cells as judged by RT-PCR and Western blotting; the expression level of Src in inducible cells was the same as those in uninducible cells, but activity of Src kinase in inducible cells was higher than those in uninducible cells.The level of phosphated tyrosine in Src was reduced in inducible cells. The malignant changes of phenotypes in the inducible cells were verified by flow cytometry and electron microscope. The results show that 24 h induction of PTPalpha could initiate the transformation of NIH3T3 cells transfected with PTPalpha inducible expression vectors, and the transformation may be associated with activated Src where pTyr(527) in the C tail was dephosphorylated by increased expression level of PTPalpha in the cells.

1276. [Extraction of the salt-tolerant cDNA in mangrove Avicennia marina by mRNA differential display].
RNAs of the leaves in Avicennia marina, which cultured in 50@1000 and 0@1000 salinity condition respectively, were isolated for mRNA differential display analysis. Screened by OligodT12 GC and eight 10-oligonucleotide arbitrary primers, differential cDNA fragments-csrg1(600 bp), csrg2(550 bp), csrg3(480 bp), only appeared in the leaves of Avicennia marina cultured in 50@1000 salinity condition. After detected by RNA dot hybridization, only csrg1 appeared the difference between the RNAs of the leaves in Avicennia marina cultured in 50@1000 and 0@1000 salinity condition respectively, and csrg1 was confirmed as the salt-tolerant cDNA. csrg1 was cloned and sequenced. After searching in Genbank, there were no similar sequences reported.

1280. [Studies on molecular teratogenic mechanism of methylmercury in early developing rat embryos].
The present study further revealed the teratogenic mechanism of methylmercury chloride (MMC) during rat neurulation by means of nonradioactive in situ hybridization (ISH), semiquantitative analysis of immunohistochemical staining and in vivo teratogenic test. The main aim was to teat the hypothesis that iNOS, HSP70, NT, TGF-beta and Bcl-2 genes contribute to MMC-induced day 9.5 embryonic damages. Results showed there were no obvious poisoning signs and death of pregnant female rats injected intraperitoncally with 0, 0.2, 0.4, 0.8, 1.6 and 3.2 mg/kg MMC. While the doses increased, the total morphological scores decreased gradually, and the rates of embryo deformity and development delay increased step by step to 34% and 76% respectively. The levels of iNOS mRNA and protein and HSP70 mRNA increased, and NT mRNA with its protein became down, all these changes were concentration dependent. In addition, MMC could inhibit the level of TGF-beta mRNA, but no obvious influence on the levels of Bcl-2 mRNA and Bcl-2 protein. On the basis of parallel findings from embryos, genes and proteins, abnormal expression of genes in transcriptional level might be related to MMC-induced teratogenic insult.
