781. [Quantitative analysis of nuclear proteome in apoptosis hepatoma cells induced by hydroxycamptothecin].
The experimental foundation for investigating the pharmacological action of hydroxycamptothecin at subcellular quantitative proteomic level can be obtained depending on the information of differentially expressed nuclear proteins in hydroxycamptothecin-treated cells and control cells. The apoptosis was induced by hydroxycamptothecin in hepatoma cells, the nucleus of cells were isolated and verified with western blot. Nuclear proteins labelled with cleavable isotope-coded affinity tag (c-ICAT) reagent were digested and purified. The expression ratio of the identical nuclear protein derived from apoptosis cell and control cell can be gained using shotgun proteomic method based on multiple dimensional liquid chromatography-linear ion trap/orbitrap mass spectrometer combined with c-ICAT strategy. A total of 42 nuclear proteins were significantly (P<0.05) altered in hydroxycamptothecin-treated cells, among them, 12 proteins showed significantly down-regulation, and 30 proteins showed up-regulation compared with control cells. The function of these proteins were likely involved in life processes of cells such as proliferation, apoptosis, differentiation, energy metabolism, nucleic acid synthesis and metabolism, structure of cell skeleton.

Activator protein-1(AP-1) is an important transcription factor, and dysregulation of its activity has been associated with many human diseases, including various cancers. A novel human gene AC3-33 (GenBank name: c30rf33, Accession No. FLJ31139), which can suppress PMA and Ionomycin induced activation of AP-1, was identified from 650 human func-tion-known genes by using the high throughput-high content cell-based screening technology. The gene whose full cDNA length is 1391 bp containing 6 exons and 5 introns is located in the human chromosome 3q25.31. The predicted protein encoded by this gene contains 251 amino acids with a theoretical molecular weight of 29 kDa. Expression of the AC3-33 gene is widly found in adrenal glands and cervix. The amino acid sequences of AC3-33 is highly conserved, and has no homology to other known proteins. Subcellular localization studies show that the AC3-33 protein was localized in the cytoplasm. Our preliminary results showed that AC3-33 is an important novel gene related to supress AP-1 activity.

To search for the molecular biomarkers of sub-lethal polycyclic aromatic hydrocarbons (PAHs)-contamination of soil, the subtractive cDNA libraries of earthworm Eisenia fetida exposed to benzo(a)pyrene (BaP) in artificial soil were constructed by suppression subtractive hybridization. After sequencing and analyzing with basic local alignment search tool (BLAST), two clones matching heat shock protein 70 k Da (HSP70) and one clone matching heat shock protein 90 k Da (HSP90) were isolated from the up-regulated library, and subsequently, the up-regulation of HSP70 and HSP90 was verified by real-time PCR in E. fetida exposed to 0.1 mg x kg(-1) and 1.0 mg x kg(-1) BaP. It was indicated that these two newly identified HSPs in E. fetida were the potential molecular biomarkers for soil contamination monitoring.

To investigate the feasibility of using exogenous salicylic acid (SA) to regulate the allelopathic weed suppression of rice, this paper studied the effects of different concentrations exogenous SA on the weed-suppression and physiological-biochemical characteristics of allelopathic rice PI312777, and the relative expression quantity of gene ZB8 in the rice by real time quantitative RT-PCR (FQ-PCR). The results showed that exogenous SA induced the enhancement of the allelopathic potential of donor rice in suppressing target weed plants, and this inducing effect was SA dose and treatment time dependent. After foliar spraying SA, the inhibitory effect of PI312777 on barnyardgrass increased significantly, and the root vigor and superoxide dismutase (SOD) and peroxidase (POD) activities of PI312777 increased, while its catalase (CAT) activity decreased. The reverse was true for the target weed barnyardgrass. At the same time, the phenylalanine ammonialyase (PAL) activity of PI312777 increased significantly, leading to an increase of the total content of phenols. Among the test concentrations of SA, 0.2 mmol x L(-1) had the best inducing effect, under which, the relative expression quantity of ZB8 increased first and decreased then, with a peak at 24 hours after treatment.

800. [Overexpression of cotton, vacuolar H+-ATPase subunit C promotes cell elongation and regulates stress tolerance in fission yeast].
Cotton (Gossypium hirsuturm L.) vacuolar H+-ATPase plays an important role in fiber cell elongation through regulating the turgor pressure, which is involved in polarity enlargement of fiber cell. Arabidopsis vacuolar H+-ATPase subunit C (DET3) regulates the activity of vacuolar H+-ATPase and subsequently promotes cell elongation. To rapidly characterize the function of cotton vacuolar H+-ATPase subunit C (GhDET3) and speculate the role of GhDET3 in cotton fiber growth and development, we constructed an expressing vector for fission yeast and introduced it into fission yeast. The results revealed that overexpression of GhDET3 was capable of promoting fission yeast cell elongation and enhancing the tolerance to high NaCl and high pH stresses. These results illuminated that the effects of GhDET3 were important for the activity of vacuolar H+-ATPase and suggested that GhDET3 plays a crucial role in fiber cell elongation.
