The protective effect of Cu, Zn, SOD on bone marrow hematopoietic progenitor cells of mice, under 4 degrees C storage was studied. The results showed, if 1.65 U or 0.165 U/ml SOD was added to the cell suspension for 3 days at 4 degrees C, the production rate of CFU-GM, CFU-E, BFU-E, CFU-Meg and CFU-Mix were 6.2, 2.6, 2.9, 4.0 and 5.1 times that of control group (without SOD) respectively. The survival rate of hematopoietic progenitor cells increased obviously. The mechanism of SOD effect is supposed to prevent hematopoietic progenitor cells from death, instead of the regulating proliferation. It may be closely related to the antioxidation of SOD which scavenges the superoxide free radicals.

The effects of LiCl on the proliferation of high proliferative potential colony-forming cells (HPP-CFC) and granulocyte macrophage colony-forming unit (CFU-GM) from bone marrow of BALB/c mice were observed. The colonies of HPP-CFC were supported by IL-1, IL-6, WEHI-3 conditioned medium (WEHI-3 CM, which contained IL-3) and L929 conditioned medium (L929-CM, which contained M-CSF); the colonies of CFU-GM were supported by WEHI-3 CM. It was shown that LiCl at concentrations ranging from 0.4 mmol/L to 2 mmol/L significantly inhibited the proliferation of HPP-CFC (P < 0.01), but concentrations ranging from 0.4 mmol/L to 1 mmol/L significantly increased the production of CFU-GM (P < 0.05), both effects displayed in dose-dependent manners. The different effects of LiCl on the proliferation of HPP-CFC and CFU-GM suggest that LiCl may induce HPP-CFC differentiation into more mature cells.

Substance P (SP)-immunoreactive cells and the axon terminals are widely distributed in the central amygdaloid nucleus (AC) and its important projection areas. The present study showed that (1) Excitation of the AC by glutamate (Glu) or injection of SP into the AC projection areas: locus coeruleus (LC), nucleus parabrachialis (NPB), periaqueductal gray matter (PAG) or lateral hypothalamus-perifornical region (LH/PF), all elicited pressor response. (2) Preinjection of DPDPDT (a SP antagonist) into bilateral LC, NPB, PAG or LH/PF could attenuate the AC pressor response to Glu. (3) Intra-RVLM (rostral ventrolateral medulla) preinjection of either phentolamine, propranolol or atropine (but not GDEE, a Glu antagonist) could also reduce the AC pressor response. Taken together with our previous findings that the alpha-, beta-, M-receptors in RVLM mediated the pressor response to LC excitation, alpha-receptors mediated the NPB pressor response, alpha- and beta-receptors mediated the PAG pressor response; these results indicate that the SPergic projections of the AC not only directly act upon the brainstem pressor areas (LC, NPB, PAG)-RVLM system, but also indirectly via the LH/PF act upon the brainstem pressor areas-RVLM system to induce the pressor response.

By using Y chromosome specific sex-determining region (Sry) as a new cytogenetic marker and PCR technique, the characteristics of proliferation and differentiation of hematopoietic stem cells in mice were studied. Bone marrow cells from male mice were injected into lethally-irradiated female mice, PCR results indicated that all of the CFU-S were originated from donor. It was found that CFU-S was able to proliferate and differentiate into various hematopoietic cells in vivo during its transplantation. Whereas the fibroblasts within donor CFU-S and the fibroblasts from bone marrow in recipient mouse reestablished by donor CFU-S were shown to be originated from the recipient. The above data demonstrated that CFC-S from bone marrow in mice possessed the characteristics of hematopoietic stem cells but was not prone to differentiate into fibroblasts.
