703. [Study on the progress of the mechanism of TGF-beta in the scarring and the effect of TCM].
Transforming growth factor (TGF-beta) plays a central role in wound healing and scarring. TGF-beta is a member of cytokine family, involving cell proliferation, migration, differentiation and apoptosis, which subsequently trigger extracellular matrix (ECM) deposite and collagen overproduction. A number of regulators of TGF-beta expression bave been identified, including Decorin CTGF, etc. TCM can reduced later scarring, and it accelerates wound healing. TCM can reduce the expression of TGF-beta, resistance of fibrosis, so that lighten the development of scar. This article reviewed the mechanism of TGF-beta in the scarring and TCM can decrease the development of scar effected by TGF-beta.

Gene PsPIP1 (GenBank accession No. EU626398) containing a complete ORF was obtained using rapid amplification of cDNA ends (RACE) from the cDNA library of Polygonum sibiricum Laxm. leaves. The length of cDNA was 1 004 bp, which encoded a peptide of 285 amino acid residues. Based on other kinds of plant aquaporin amino acid sequences, the phylogenetic evolution, and tertiary structure of protein comparison, this gene was classified into aquaporin subfamily. Expression analysis by Real-time PCR showed that PsPIP1 gene was expressed in leaves, stems, and underground stems. The expression level of PsPIP1 gene was higher in leaves than in underground stems and was the lowest in stems. The expression pattern of PsPIP1 gene induced by NaHCO3 stress and de-stressing also varied remarkably.
715. [Expression profile analysis of Peptide5 and Peptide6 in Arabidopsis].
Two predicted peptide genes in Arabidopsis thaliana L., Peptide5 and Peptide6, was confirmed by RT-PCR in mRNA level. The expression profile indicated that both genes were generally expressed at different developmental stages and tissues as constitutive gene expression, and they also responded to six treatments including NaCl, PEG, MeJA (methyl jasmonate), SA (salicylic acid), cold and wound in transcription level. Analysis of the promoter sequence suggests that Peptide5 in Arabidopsis may contribute to the secondary xylem formation.
716. [Cloning and expression analysis of two salt and Fusarium graminearum stress associated UDP-glucosyltransferases genes in wheat].
Glycosyltransferases (GTs) play important roles in stress responses of plants by glycosylating hormones and secondary metabolites. UDP-glucosyltransferases (UGTs), which use UDP-glucuronic acid in animals, UDP-glucose, UDP-galactose, and UDP- rhamnose in plant as sugar donors, belong to family 1 of GTs. As a secondary metabolite produced by Fusarium graminearum during infection of grains, deoxynivalenol (DON) is not only harmful to human and animal's health by inhibiting protein synthesis, but also acts as a virulence factor during fungal pathogenesis. In order to study expression profile of UGT genes in wheat, two UGTs, designated TaUGT1 and TaUGT2, were isolated from bread wheat (Triticum aestivum L.) by reverse transcriptase-polymerase chain reaction. The genomic sequences of both genes had one intron. Their coding sequences shared 91% and 90% similarities at nucleic acid level and the deduced protein sequence level. The analysis of conserved domain revealed that these two cDNAs encoded UDP-glucoronosyl and UDP-glucosyl transferase with PSPG (Putative secondary plant glycosyltransferase) domain. Real-time PCR was carried out to detect the expression profiles of the two UGTs in wheat under various stress conditions. In young spikes infected by Fusarium graminearum, TaUGT2 was induced but TaUGT1 was repressed. These two genes were upregulated under higher NaCl concentration. In conclusion, TaUGT2 may participate in wheat resistance to Fusarium head blight in which mycotoxin DON plays a role, and these two genes might be involved in responses of wheat to salt stress.

S100A4 is an important metastasis-associated gene. Researches have confirmed the close correlation between overexpression of S100A4 gene and gastric cancer's infiltration, lymph node metastasis and in vitro invasiveness of gastric cancer cells. In order to investigate the mechanism of overexpression of S100A4 gene, hypoxia mimetic cobalt chloride (CoCl2) was used to treat gastric cancer cell BGC823, and then the expression of S100A4 mRNA and protein in BGC823 cells were detected by RT-PCR, immunohistochemistry, immunofluorescence, and Western blotting analysis. After treatment with CoCl2, the expression of S100A4 mRNA and protein in BGC823 cell was increased. These results suggested that hypoxia mimetic cobalt chloride could increase the expression of S100A4 gene in gastric cancer cell BGC823.
