Observations on structure and distribution of vimentin as determined by indirect immunofluorescence were showed that the arrangement of vimentin intermediate filments (IFs) changed from an extended fibrillar state to a complex cage formation tightly associated with the forming lipid globules during adipose conversion of rat preadipocytes. The effects of differentiation of rat preadipocytes into adipocytes on vimentin mRNA levels as determined by in situ hybridization (ISH) using oligonucleotides probes to vimentin mRNA labeled by digoxigenin (DIG), and on vimentim protein expression as determined by Western blotting were researched. The results suggested that the expressions of vimentin with molecular weight of about 57 kD at the levels of both mRNA and protein was existed throughout the predipocyte differentiation and downregulation of vimentin is required for adipose conversion. The specificity of the association of lipid globules with vimentin IFs during adipose conversion was taken as an example of a possible IF function in relation to a preadipocytes differentiation process, especially discussed as a functional relationship in supporting adiposgenesis.
6602. [Progress in the studies of parthenogenetic embryonic stem cells].
Human embryonic stem cells can be induced to differentiate into all kinds of cells in vitro to be applied to clinical medicine and scientific research. Because they have to be isolated from human embryos, any attempt to establish human stem cell line is prohibited by religion and ethics in some areas and countries. Parthogenetic embryonic stem cells have similar capacity of totipotency and proliferation, and can be established from parthenogenetic activation of discarded oocytes. This paper reviews the progress in the studies of the establishment of parthenogenetic embryonic stem cell line and its differentiation capacities.

Systematic lupus erythematosis (SLE) is a severe disease which affects the patient for many years and there is no radical cure for the disease. To explore a possible way to treat children with refractory SLE, the authors treated 2 children with grade III and IV lupus nephropathy for 5 years and 7 years respectively, mainly presented with persistent thrombocytopenia, proteinuria, pleural effusion with CD34(+) autologous peripheral progenitor cells transplantation.

Mesenchymal stem cells (MSCs) were adult stem cells which contribute to the regeneration of mesenchymal tissues such as bone cartilage, muscle, ligament, tendon, adipose and stroma. Due to the multipotential ability and self-renewal capacity, the mesenchymal stem cells can be applied in many fields, such as the seed cells in tissues engineering, cell therapy and gene therapy. To enhance the clinical use of MSCs, the investigators studied the isolation and expansion of MSCs from adult bone marrow, fetal bone marrow and human umbilical cord blood, and investigated their biological identities.
