A population of F8 recombinant inbred lines (RILs), derived from a cross between IR26 (Oryza sativa L. spp. indica) and Jiucaiqing (japonica), were used to identify the quantitative trait loci (QTLs) for Cd2+ content in brown rice under 5 mg/kg Cd2+ stress. Two QTLs, qCCBR-11a and qCCBR-11b, associated with the Cd2+ content in brown rice, were detected on chromosome 11. qCCBR-11a was located at the position between markers RM6288 and RM6544, accounting for 11.17% of the phenotypic variance with an additive effect value of 0.089. qCCBR-11b at the interval between markers RM167 and RM5704 explained 7.66% of the phenotypic variance with an additive effect value of 0.075. In addition, the correlation coefficients between Cd2+ content of brown rice and plant height, spikelets per panicle, filled grains per panicle, seed setting rate, and 1 000-grain weight were not significant. This suggested that the Cd2+ content in brown rice under Cd2+ stress was an independent genetic trait.

This study was purposed to investigate the changes of apoptosis-related gene expression in T lymphocytic leukemia JM cells induced with matrine, and its possible mechanism. JM cells was induced with 0.4 mg/ml matrine for 4 days, the total RNA was extracted from JM cells before and after matrine induction, the differential expression of apoptosis-related genes were screened with cDNA Expression Array Kit, the expression change of a part of gene was checked by Western blot. The results indicated that after induction of JM cells with matrine, differential expression of 31 genes were found by gene chip hybridization, the expression of caspase 8 was up-regulated more than 5 times. Western blot analysis showed that the up-regulation of caspase 8 gene expression positively correlated with induction time. It is concluded that differential expressions of many apoptosis-related genes in JM cells can be induced by matrine, in which gene expression of caspase 8 is up-regulated notably.

The mechanism of Tripterygium wilfordii (GTW) inhibiting productive function was studied in the male mice. The male mice were randomly divided into control group and GTW group. GTW group had been given Tripterygium wilfordii for 8 weeks. The pregnancy rate was investigated after the male mice mated with the normal female mice (1ratio2) in one week. Also the pathological changes of the male mice testis were evaluated by the optical microscope and the expression of genes related to reproduction was investigated by Gene Chip Technology 8 weeks later. The results showed pregnancy rate of GTW group was obviously decreased compared with the control mice, and no pregnancy occurred in GTW group at 8 weeks. In the testis tissues of GTW group, the spermatogenic cells in the convoluted seminiferous tubules were obviously decreased and the lumen was obstructed by large deciduous spermatogenic cells. The number of abnormal expression genes in this group was 1 932. Among them, 354 genes were related to reproduction, including 112 up-regulated and 242 down-regulated genes. Genes Herc4, Ipo11, and Mrto4, which are closely associated with spermiogenesis, were expressed irregularly. The outcome indicates that the inhibitive reproduction of Tripterygium wilfordii in the male mice is associated with the abnormal expression of spermiogenesis genes.

The induction and measurement of cytochrome P450 in white rot fungus Phanerochaete chrysosporium were studied in this work. The spectrophotometric results demonstrated that n-hexane was able to induce the fungal P450 to high level, which facilitated isolation and measurement of microsomal P450. The highest concentration of microsomal P450 could reach 140-160 pmol/mg after 6-h-induction by addition of 2 microL/mL hexane each hour, and the concentration of hexane and incubation time had significant effect on the induction of P450s. After effective induction, the method for isolation and measurement of microsomal P450 with CO difference spectrum was studied and the optimized method was obtained as followed. High-speed disperser and glass homogenizer were used to disrupt cells, which obtained higher amount of microsomal P450 than those from cells disrupted by glass homogenizer, ultrasonicator and bead-beater respectively. To record CO difference spectrum,the sample was bubbled with CO for 40 s at a rate of 3 mL/min (300 microL sample), and the reference cuvette was bubbled with N2 to the same extent. Then, the reducer sodium dithionite was added to a concentration 0.4 mol/L.
