The prognosis of relapsed or refractory aggressive non-Hodgkin's lymphoma (NHL) after front-line therapy remains poor. The development of more effective and less toxic salvage regimens remains a major challenge. Gemcitabine is effective in treating lymphoma and, when combined with cisplatin, is effective for some solid tumors. This study was to evaluate the efficacy of GDP regimen (gemcitabine, dexamethasone, and cisplatin) on relapsed or refractory aggressive NHL, and observe the adverse events.

The developmental potential of reconstructed embryos varied according to the source of donor cells, it was thought that the donor cells capabilities to be reprogrammed were different. We established the method of culturing porcine bone marrow mesenchymal stem cells (pMSCs), identified and observed the growth characteristics of pMSCs, and determined pMSCs reprogramming potential as donor cells for nuclear transfer (SCNT). We found that the method of gradient centrifugation to isolate pMSCs from porcine bone marrow was better than the method of anchoring culture; the number of pMSCs achieved peak at day 6, the adhesive rate of cultured cells was 78.50% at 10h and the division index of cultured cells was 24.00 per thousand at day 4. The developmental competence were compared among three kinds of embryos, reconstructed embryos with PF and pMSCs, Parthenogenetic. The blastocysts rate and total cell number of blastocysts were 15.07%, 14.63% vs 30.91% and 24.1 +/- 6.5, 30.67 +/- 17.7 vs 25.8 +/- 11.4. These results indicated that pMSCs could be high proliferation and stable growth characters in vitro, and were suitable donor cells type for nuclear transfer.

As a concentrated location for adult somatic stem cells, the niche plays a crucial role on maintaining the high proliferative ability and the inducing the particular differentiation of the stem cells by providing the special microenvironment containing definite extracellular matrix and niche cells. Nowadays limbal stem cells (LSCs) are the acknowledged adult somatic stem cells of corneal epithelium. Rapid progress has been accomplished in the theory and clinical practice on LSCs. However, the understanding on its niche has obviously fallen behind. This review summarized and analyzed the status quo of the study on niche of LSCs, mainly through the following aspects: the precise location of the niche was described as limbal epithelial crypts; the blood-derived and stroma-derived cytokine could regulate the proliferation, differentiation and apoptosis of LSCs by triggering particular intracellular information pathway; the survival and function of LSCs could be influenced by damaged niche microenvironment, while the differentiation of LSCs could be veered by another type of niche; in ex vivo LSCs expansion, reconstruction of the niche is needed and could be achieved by 3T3 cells or using amniotic membrane as the carrier.

Although the drugs, such as Memantine, Calpain, Erythropoietin, have demonstrated exciting results for neuroprotection in laboratories, the phase III clinical trial of Memantine failed to prove such activity. So far, none of neuroprotection drugs has been approved by FDA for clinical use with the failure of Memantine clinical trail indicating that the gap between basic science research and clinical application in glaucomatous optic neuroprotection remains to be filled. This paper offers a new insight into the field of neuroprotection in glaucoma. To make the dream of optic neuroprotection to reality, we have to implement new perspective strategies to integrate technologies and findings from the researches of human genomics, proteomics, stem cells, and gene-transferred animal models.

Non-small cell lung cancer (NSCLC) is now regarded as the most common cause of cancer-related mortality in China. Despite continuous efforts to improve the therapeutic response, the overall five-year survival rate for NSCLC is still less than 15%. Now we have known that the growth of neoplastic tumors is maintained exclusively by a small subpopulation called "cancer stem cells" which posseses ability of self-renew and differentiation. It has been widely accepted that cancer stem cells are chemoresistant and radioresistant. Therefore, a major challenge in treating this and other cancers is the intrinsic resistance to conventional therapies demonstrated by the stem/progenitor cell that is responsible for the sustained growth, survival, and invasion of the tumor. Identifying these stem cells in non-small cell lung cancer and defining the biologic processes necessary for their existence are paramount in developing new clinical approaches with the goal of preventing disease recurrence. This review summarizes our update understandings of the cellular and molecular mechanisms operating within the putative cancer-initiating cells at the core of non-small cell lung cancer.
