Stem cells and miRNAs are two of the most intriguing research topics in recent years. Stem cells with properties of self-renewal and multipotency are remarkable cells since they play key roles in animal development and tumorigenesis. microRNAs (miRNAs) are approximately 22nt non-coding RNAs, which have spatiotemporally specific expression patterns, and are highly conserved between species. Researchers have demonstrated that miRNAs can regulate gene expression at post-transcriptional level, and their action is one of the essential intracellular regulatory pathways. Recent evidence showed that miRNAs can regulate stem cells self-renewal and differentiation. The evidence mostly derived from the following research strategies: (1) deletion or mutation of the genes that encoding essential enzymes for miRNAs biogenesis, such as Dicer1, Loqs, DGCR8, and Argonaute; (2) screening of specific miRNAs from stem cells and investigation of their function. Comprehension of the miRNAs functions in stem cells is of great importance for further understanding their self-renewal and differentiation mechanism, as well as identification of stem cells. This paper reviews the potential roles of miRNAs in stem cells on the basis of recent researches.

Aorta-gonad-mesonephros (AGM) is well known as a main structure that de novo generates hematopoietic primary stem cells (HSC) in mid-gestation mammalian embryos. Hemogenic endothelium, and recently, subendothelial mesenchyme as well as hemangioblast are shown as contributing to blood formation in AGM region. AGM-HSC displays dynamic changes in surface markers, including CD41, CD45 and several endothelial-specific molecules. The novel finding of interleukin-3 as a potent regulator of AGM-HSC seems very interesting. Moreover, zebra fish model reveals PGE2 as a novel stimulator of HSC in AGM and kidney marrow, which is also the case in mouse hematopoietic tissues. Identification of mesenchymal stem cells with significant hematopoietic supporting capacity in AGM region suggests an alternative pathway to explore new molecules governing embryonic and adult hematopoiesis. In this paper, the hemogenic model in AGM region, surface markers on HSCs in AGM region and regulation of HSCs in AGM region were reviewed.

Matrix metalloproteinases (MMPs) have the ability to degrade extracellular matrix components. They abnormally express in a variety of solid tumors and hematologic malignancies, and play an important role in tumor invasion and metastasis through extracellular matrix degradation, which closely relates with the progression and prognosis of the malignant disease. This article reviews progress of study on the mechanism of MMP underlying the pathogenesis of hematologic malignancies, including structure of MMP, regulation mechanism of MMP and its relation with proliferation and differentiation of hematopoietic stem cells (HSC), angiogenesis and tumor immunology and so on.
