Anti-synthetase syndrome (ASyS) is a rare autoimmune disorder defined by anti-synthetase antibodies. While malignancy is established in DM, its association with ASyS remains unclear. We aimed to estimate malignancy prevalence in ASyS and assess whether autoantibody subtype, age, sex or clinical features influence malignancy risk.

Takayasu arteritis is a rare vasculitis characterized by inflammation of large arteries. Given the robust genetic association with HLA-B*52:01, it was recently proposed to classify Takayasu arteritis within the spectrum of MHC-I-opathies. Although genetic associations in ERAP1 and ERAP2 have been described in several MHC-I-opathies, their role in Takayasu arteritis remains unclear. This study investigated the genetic interaction between ERAP1/ERAP2 and HLA-B*52:01 in Takayasu arteritis.

Anti-RNA polymerase III antibodies (ARA) are frequent in systemic sclerosis (SSc). However, the reported prevalence is variable among studies and some clinical associations are debated. We aimed (i) to update the recent data on overall ARA prevalence in SSc and heterogeneity between centres; and (ii) to describe their clinical associations.

The ASAS consensus defines 'early axial spondyloarthritis (axSpA)' as symptom duration ≤2 years, a definition derived from expert opinion due to limited evidence. We performed a meta-analysis of randomized placebo-controlled trials (RCTs) of biologic and targeted synthetic DMARDs (bDMARDs/tsDMARDs) in axSpA to assess the impact of symptom duration on treatment response.

ANCA-associated vasculitis (AAV) is a group of systemic autoimmune diseases affecting small blood-vessels. Class-II human leukocyte antigen (HLA) genes are often reported as major genetic determinants. We conducted a systematic review and meta-analysis to evaluate the susceptibility conferred by HLA genes to AAV and five of its clinical subtypes [PR3+AAV, MPO+AAV, granulomatosis with polyangiitis (GPA), microscopic polyangiitis (MPA) and eosinophilic granulomatosis with polyangiitis (EGPA)].

IBM is characterized by slowly progressive muscle weakness making it challenging to detect significant changes in weakness during a clinical trial. Trial participants receiving placebo may behave differently from in natural history studies. We aimed to quantify the change in muscle strength and IBM Functional Rating Scale (IBMFRS) of IBM patients receiving placebo during clinical trials.

Among adults who develop coronavirus disease 2019 (COVID-19), those with rheumatic diseases (RDs) have similar hospitalization rates compared with those without RDs. Similar comparisons are lacking in children, due to the overall rarity of COVID-19-related hospitalization in this population. We aimed to examine the risk factors for COVID-19-related hospitalization in paediatric patients with RDs.
